An Antiserum to PIG IL-1 (Catabolin) Reacts with the Acidic But Not the Neutral Form of Human IL-1

J. Saklatvala, S.J. Sarsfield and D.D. Wood

   Abstract

An antiscrum was made in a rabbit against pig IL-1 (catabolin) The antiscrum was shown by Western blot to react with the antigen and also to cross-react with certain other slightly larger leucocyte proteins The antiserum directly inhibited the action of pig IL-1 in a mouse thvmocytc assay and also prevented it from inducing rcsorption of proteoglycan in explants of bovine nasal cartilage The antiserum inhibited partially purified human IL-1 of pl 5.0 m both the thymocyte and cartilage assays, but it did not interact with the human IL-1 of pl 7.0 The results suggest that pig IL-1 (catabolin) and human IL-1 pl 5.0 may be related and that the human IL-lsare immunologically different.

KEY WORDS: IL-1, Catabolin, Cartilage, Proteoglycan
